Abstract

INTRODUCTION: High failure rates in clinical drug development are often attributed to inadequate therapeutic efficacy. While binding affinity has traditionally guided lead optimization, it reflects only the equilibrium state of drug-target interactions and often correlates poorly with in vivo pharmacological responses.
